ISOLASI DAN KARAKTERISASI SENYAWA METABOLIT SEKUNDER DARI FRAKSI AKTIF DAUN TEMPUYUNG (Sonchus arvensis L.) TERHADAP UJI TOKSISITAS

Abstract: Isolation  of  secondary  metabolite  compound  from  tempuyung  leaves  (Sonchus  arvensis  L.)  has been  done.  Toxicity  assay  of  n-hexane  extract,  ethyl  acetate  extract,  methanol  extract  and  water extract has been tested, and methanol extract obtained high activity in toxicity assay using Brine Shrimps  Lethality  Test  (BSLT)  with  LC50  values  403.645  mg/L.  Isolation  was  continued  by maceration  using  methanol  solvent.  Methanol  extract  was  soluted  with  water  and  than  was fractionated  with  n-hexane  and  has  been  continued  by  ethyl  acetate  solvent.  Toxicity  of  each fractions was tested, and water fraction obtained highest activity with LC50 values 269.774 mg/L. Water fraction was freeze-dried and column chromatographied using silica gel  as the stationary phase  and  n-hexane,  ethyl  acetate  and  methanol  as  the  mobile  phase  with  using  Step  Gradient Polarity  (SGP)  method.  Isolated  compound  is  white-brown  solid  around  3  mg  which  has  single spot  with  some  eluent  comparisons  on  thin  layer  chromatography.  The  results  of  chemical characterization  of  isolated  compound  using  spectroscopy  method  indicates  that  the  isolated compound was included as coumarin group.
