ISOLASI DAN KARAKTERISASI SENYAWA KUMARIN DARI KULIT BATANG Fagraea ceilanica Thunb. SERTA UJI ANTIOKSIDAN

Abstract: Isolation  of  coumarin  compounds  from  the  bark  of  Fagraea  ceilanica  Thunb.  have done  by maceration method using a solvent of n-hexane, ethyl acetate and methanol. A silica gel column was  used  for  the  chromatographic  isolation  of  the  ethyl  acetate  extract  components.  N-hexane and ethyl acetate as the mobile phase in the SGP (Step Gradient Polarity). Isolated compounds in the form of dark yellow crystals as much as 26 mg which has a melting point of 205 - 207ÂșC. It gave a single spot in TLC (Thin Layer Chromatography) (Rf 0.42) which eludated with mixture of  n-hexane  :  ethyl  acetate  (7:3).  It  fluorescent  under  365  nm  UV  and  become  brighter  after sprayed with 5% NaOH as the specific reagent for coumarin. Based on the UV and IR spectrum of  this  compound  showed  absorption  chromophore  lactone  ring  and  has  a  functional  group  -OH at 3318 cm-1, C-H stretching at 2926 cm-1, C = O stretching at 1698 cm-1, C = C stretching at 1602 cm-1, and C-O stretching at 1263 cm-1 which indicates that the isolated compounds such as coumarin  group  which  has  the  -OH  group.  Test  results  antioxidants  with  DPPH  free  radical method  shows  methanol  and  ethyl  acetate  extracts  were  moderate  to  its  activity  as  an antioxidant which has IC50 values respectively - also at 118.76 mg/L and 175.68 mg/L, whereas the  n-hexane  extract  inactive  with  IC50  value  of  6524.67  mg/L.  Isolated  compounds  also  has potential as a relatively weak antioxidant with IC50 value of 358.71 mg/L.
