ISOLASI DAN KARAKTERISASI SENYAWA TRITERPENOID SERTA UJI ANTIBAKTERI PADA DAUN Ficus variegata Blume

Abstract: Isolation of triterpenoid compound from the leaves of Ficus variegata Blume have been done by maceration  method  using  n-hexane  solvent.  N-hexane  extract  was  chromatographed  column using  a  silica  gel  as  the  static  phase,  n-hexane  and  ethyl  acetate  8  :  2  as  the  mobile  phase  in  a Isocratic  manner. Isolated  compounds resemble  the  white needle crystals  with  a melting point in  136º  -  138ºC.  TLC  test  (Thin  Layer  Chromatography)  gave  a  purple  single  stain  using Liebermann-Burchard reagent with Rf 0.5 which not fluorescence under UV rays at λ 254 nm and 365  nm.  Based  on  UV  and  IR  spectrum  this  compound  indicates    presence  of  double  bond which  not  conjugated  at  212,14  nm  and  having  a  functional  group  –OH  at  3439,39  cm-1,  C-H stretching  at  2935,69  cm-1,  C=O  stretching  at  1710,57  cm-1,  C=C  stretching  at  1638.34  cm-1,  C-O stretching at 1051.83 cm-1, 1376.76 cm-1 and 1466.05 cm-1  are geminal dimethyl group (C-(CH3)2) which indicates that the isolated compound  is triterpenoid. The result of antibacterial test using diffusion  test  in  order  to  show  medium  active  of  n-hexane  extract  at  concentration  20  mg/mL with hallozone 7.32 mm as antibacterial against bacteria of  Staphylococcus aureus and not active in Escherichia coli bacteria.
