Abstract: Isolation and
characterisation of alkaloid
compound from antibacterial active
fraction of betel leaves
(Piper betle Linn)
have been done.
Maseration of Piper
betle leaves using
hexane, ethyl acetate and
methanol obtained 47.653
g heksane extract,
62.225 g ethyl
acetate extract, and 75.270 g
methanol extract. These
fractions were determined
their antibacterial activities
agains pathogenic toward Gram positif and Gram negatif bacteria, Staphylococcus aureus and Escherichia coli, using
disc diffusion method.
Ethyl acetate fraction
was the greatest
antibacterial activity. This
fraction separated with column chromatography by Step Gradient Polarity (SGP)
with silika gel 60 F254 as adsorbent and
hexane : ethyl acetate as eluents. The result was to be 50 g of alkaloid campound
as white needle crystal which showed orange spot using Dragendroff reagent on
TLC plate. The alkaloid
observed in this
work melted in
159,1-161,1oC. The ultraviolet
spectrum showed maximum absorbtion
in 264,80 nm.
IR spectrum showed
vibrations in 3544,04
cm-1, 2968,32 cm-1 and
2937,19 cm-1, 1747,50
cm-1 and 1702,14
cm-1,1623,34 cm-1 and
1511,47 cm-1, and 1051,99 cm-1 with function groups -NH,
C-H steching, C=O, C=C aromatic, and C-N streaching.
