PRODUKSI SELULASE DARI Aspergillus niger DAN KEMAMPUANNYA MENGHIDROLISIS AMPAS TEBU
Abstract: Cellulase enzymes
has been produced by A. niger and used for the hydrolysis of bagasse. The
enzyme produced by inoculate the fungus A. niger in to fermentation media with
each contained inducer CMC, and inducer sugancane baggase. Fermentation is done
by variation of 3, 6, and 9 days. After that the tested enzyme activity
generated by the CMC substrate. Glucose formed then measured with a
spectrophotometer with a wavelength of 580 nm. From the test results of enzyme
activity with the highest CMC activity obtained at optimal fermentation for 6
days of bagasse inducer. This shows that bagasse without pretreatment induces
the release of both enzymes. Cellulase activity obtained at 11.79 units. Then
do the bagasse that was pretreatment with NaOH 5%. Hidrolysis done with varying
concentrations of bagasse and bagasse hydrolysis time. The maximum glucose
concentration bagasse generated at 3% and hydrolysis time of 60 minutes is
equal to 286.25 mg / L by using enzyme extract obtained with inducer bagasse
that was pretreatment.
