PRODUKSI ENZIM SELULASE DARI Aspergillus niger DAN KEMAMPUANNYA MENGHIDROLISIS JERAMI PADI
Abstract: Cellulose enzyme
produced by Aspergillus niger has been
tested and applied in hydrolysis of rice straw. The enzyme was found to be
carboxy methyl ceulose (CMC) as carbon source, pretreatment and unpretreatment
rice straw with NaOH for 3, 6 and 9 days
of periodes. Rice straw hydrolisis was done by varying the substrate
concentration, which were 1, 2, 3, 4 and 5 %, hydrolisis time: 30, 45, 60, 75
and 90 minutes. Formed glucose was determined by Somogy Nelson method. The
highest enzyme activity was in pretreatment rice straw as carbon source, with 6
days of fermentation. This fact indicated that the pretreatment of rice straw
produced the highest of celulose enzyme with the activity of celulose was 2.10
unit with 3 % of CMC. The maximum ability of celulose enzyme in hydrolisis of
the pretreatment rice straw was 4 % in 75 minutes of hydrolisis time producing
reduced sugar to be 146.24 mg/L.
