Pengaruh Antikoagulan dan Waktu Penyimpanan terhadap Profil Hematologis Tikus (Rattus norvegicus Berkenhout, 1769) Galur Wistar
Abstract: Blood is important
component since it reflects individual physiological condition. Therefore,
blood becomes one of fundamental parameters in preclinical/biomedical test. Hematology
is the study of peripheral blood cells condition in normal and disease. Erythrocytes
and leukocytes profiles are main parameters in routine hematological evaluation.
Blood samples sometimes cannot be examined immediately for various reasons. To
keep the condition, anticoagulant is added and then stored in refrigerator for several
hours to several days. This research was carried out to study the profiles of erythrocytes
and leukocytes in healthy/normal Wistar rats (Rattus norvegicus Berkenhout, 1769)
blood samples with EDTA or Heparin and different storage duration (0, 6, 18,
24, and 48 hours). For further discussion, data were statistically analyzed
using two-factor ANOVA (P<0.05). Results showed that anticoagulants
significantly affected erythrocytes and leukocytes profiles (P<0.05).
Erythrocytes profile in EDTA-blood sample was lower than which with Heparin. In
contrast to the leukocytes profile, EDTA-blood sample was higher than which
with Heparin. With increasing storage duration, erythrocytes, total leukocytes,
neutrophils, and lymphocytes counts decreased significantly (P<0.05). Storage
duration did not significantly affect hematocrit value, hemoglobin
concentration, as well as monocytes, eosinophils, and basophils counts
(P>0.05). In conclusion, the best hematological examination is using plain
blood (without anticoagulant), however it must be accomplished immediately
after the sample is obtained. If it is impossible, EDTA- or Heparin-blood
samples can be used, in which the anticoagulant type must be stated in the report.
Likewise, hematological examination using anticoagulant also should be
performed soon after the sample is received.
