Abstract: Ascorbic acid
is a compound
that has antioxidant
properties. There are
many methods to determine
total antioxidant in
natural samples. In
this study, we
validate the method
to determine antioxidant content in natural samples. The method is using
Fe+3 as oxidator to oxidize antioxidant
and o-phenantrolin as
a complex reagent.
The formed complex
compound was measured by using
spectrophotometer. The parameter of validation that we got were correlation coefficient (r) = 0.9953; LoD =
1.14 x 10-5 M and LoQ = 7.59 x 10-6 M. While SDR and recovery for lime and
orange were 4.96
%; 2.93 % and 100.37
%; 98.91 %,
respectively. From this
result, we conclude, that the
method we validate can used to determine total antioxidant in natural samples.
