PENGGUNAAN SUMBER NITROGEN TERHADAP KANDUNGAN PROTEIN DAN ASAM AMINO PADA MIKROALAGA Spirulina platensis

Abstract: Through  an  experiment  process,  the  usage  of  nitrogen  source  to  protein  and  amino  acid  content in microalgae Spirulina platensis has been found. This research aimed to analyze protein and amino acid content in Spirulina platensis cutured in two mediums; medium with NaNO3 as nitrogen  source  and  urea  as  nitrogen  source,  and  to  determine  the  best  nitrogen  source  in Spirulina  platensis  growth.  Protein  extraction  performed  in  alkaline  extraction  method  for  5 hours  and  followed  by  precipitation  in  pH  3.  Amino  acid  analysis  was  performed  with Aminoacid  Analyzer.  Protein  content  determination  was  carried  out  using  Bradford  method that  shows  the  highest  protein  content  in  urea  biomass  culture  is  768.17  ppm.  Sodium  nitrate biomass  cuture  shows  the  lower  protein  content,  that  is  146.4  ppm.  The  growth  of  Spirulina platensis was done with spectrophotometer at 560 nm.  Amino acid analysis shows that NaNO3 biomass culture yield the higher amino acid content than urea, but urea biomass culture shows the best growth with the higher absorbant than NaNO3 biomass culture.
