DETECTION OF flaA GENE THERMOTOLERANT Campylobacter jejuni ISOLATED FROM CHICKEN MEAT USING POLYMERASE CHAIN REACTION
ABSTRACT: This study was aimed
to detect one of virulence factor, flagellin A (flaA) of C. jejuni on chicken
meat sold in traditional market by using Polymerase Chain Reaction (PCR)
method. Twelve chicken legs obtained from traditional markets were collected,
diluted in Broth Pepton Water, enrichment in Preston broth, and it was then
cultured on the modified charcoal cefoperazone deoxycholate agar (mCCDA).
Colonies on this medium were harvested and DNA was extracted. Supernatant from
DNA extraction was separated from pellet and used as DNA template. PCR
amplification was run in DNA thermal cycler on a total volume of 20 µl with annealing
temperature at 58ºC for 1 min. PCR products were analyzed using electrophoresis
method. The results showed two samples had a band of the 650 bp of 23S rRNA
gene, which means that chicken meat samples contain thermotolerant
Campylobacter sp. One sample had a band of the 1558 bp of flaA gene, meaning
one of C. jejuni that contaminated chicken meat samples contain virulence
factors.
