AKTIVITAS ANTIBAKTERIAL EKSTRAK DAUN TEMU KUNCI (Kaempferia pandurata) TERHADAP BAKTERI Aeromonas hydrophila

ABSTRACT: This  study  aims  to  assess  the  leaf  extract  of  temu  kunci  (Kaempferia  pandurata),  as  an antibacterial  of  A.  hydrophila.  The  experiment  was  conducted  at  the  Laboratory  of  Aquatic Microbiology, Faculty of Fisheries and Marine Sciences, University Mulawarman, which began in January to February 2014.  Concentration of leaf extracts are used in the temu kunci inhibition test order for Agar Disc Diffusion method (ADD) on the growth of bacteria A. hydrophila is 100 ppm, 200  ppm,  300  ppm,  400  ppm,  500  ppm,  600  ppm,  700  ppm,  800  ppm,  900  ppm,  1000  ppm,  1500 ppm, 2000 ppm, 2500 ppm, 3000 ppm, 3500 ppm, 4000 ppm, 4500 ppm, 5000 ppm, 5500 ppm and 6000 ppm.As a positive control is antibiotic Amoxicillin and fisiogis solution as a negative control. Ekstrak  dripped  on  paper  discs,  placed  on  the  media  that  has  overgrown  BHIA  A.  hydrophila bacteria.  Then further tested with a bacterial culture test with the extract, the bakteria count using the  Total  Plate  Count  and  Spektrofotomter.Concentration  was  used  the  best  ADD  600,  700,  800, 900 ppm, which the best inhibition zone ranged 10,5-13 mm.  The results showed leaf extracts of K. pandurata are antibacterial against A. hydrophila. In ADD the best leaf extract concentrations is 900 ppm, while in the bakterial culture countextract concentration is 600 ppm.
