ISOLASI DAN KARAKTERISASI ENZIM LIPASE DARI Aspergillus niger YANG DIGUNAKAN SEBAGAI BIOKATALIS UNTUK PEMBUATAN BIODIESEL
Abstract: This research is
purpose to determine the ability lipase enzyme from Aspergillus niger that used
to production of biodiesel with interesterification reaction by using methyl
acetate as substitution of methanol. Production of lipase has been done by
fermentation method for 24 hours. The optimum time of fermentation to produce
lipase at 12 hours. The fermentation result was sentrifugated by 4000 rpm for
20 minutes, supernatant was taken as crude lipase enzyme. The crude enzyme that
obtained was characterized to get optimum pH and optimum temperature. The
optimum pH was 6, optimum temperature was 30oC and the activity was 1,104 U/ml.
And then crude enzyme was fractionated by ammonium sulphate to increase lipase
activity. From variation of fractionation was obtained fractionation (80-100%)
as the highest activity with 1,542 U/ml.
The enzyme fractionation then conducted a preliminary test in the
production of biodiesel with interesterification reaction. The result was
analyzed with GC-MS and was obtained methyl decanoate.
