Effect of Genistein on Conjunctival Goblet Cells Density and the Expression of Interleukin-1bin Ovariectomized Rats
Abstract: The aim of this
research is to evaluate the effect of genistein on conjunctival goblet cells
density and IL-1b expression of ovariectomized(OVX) rats. Female adult Wistar
rats were subjected to bilateral OVX and divided into 3 groups of genistein
treatments (0.1 mg/kg/day,0.5 mg/kg/day, 1.0 mg/kg/day) subcutaneous (sc) and 3
groups of control, 2 groups of OVX rats without treatment as positive
control,and 1 group of normal rats as negative control. At the end of
procedures, right eyeball and eyelid was dissected. Periodic
acid-Schiffstaining was performed in histologic sections to determine the
numbers of goblet cells present. The expression of IL-1b was observedusing
immunohistochemistry. The number of goblet cells in OVX rats as positive
control was significantly decreased compared withnormal rats (p < 0.05).
There were no significant differences of goblet cells number observed between
negative control and all ofgenistein treatment groups (p > 0.05). There were
significant increase of IL-1b expression on positive control groups compared
withnormal (p < 0.05). Expression of IL-1b in treatment groups was not
significantly different compared with negative control at the doseof 0.5
mg/kg/day and 1.0 mg/kg/day (p > 0.05). The higher conjunctival goblet cells
density, the lower expression of IL-1b. Genisteinat the dose of 0.5 mg/kg/day
sc can improve conjunctival goblet cells number and reduce expression of IL-1b
of OVX rats. Goblet cellsdensity inversely correlated with expression of IL-1b
