PENGARUH KONSENTRASI SUKROSA TERHADAP BIOMASSA DAN KADAR SAPONIN KALUS GINSENG JAWA (Talinum paniculatum Gaertn.) PADA BERBAGAI WAKTU KULTUR
ABSTRACT: The aim of this
research was to determine the effect of sucrose concentration on callus biomass
and saponin levels of ginseng java (Talinum paniculatum Gaertn.) in various
culture of predetermined time. The research was conducted by inducing the callus
on MS medium given growth regulator 2,4-D 2 mg/L kinetin and 1 mg/L and sucrose
levels variation different at 15 g /L, 30 g /L (control) , 60 g /L, and 90 g
/L. Callus was grown
in a culture
that is different
in each sucrose
treatment. Given culture time was
4 weeks, 6 weeks and 8 weeks. The parameters that were measured include fresh
weight, dry weight and the levels of saponins. Levels of saponins were obtained from
the calculation of the area
of the stain
on silica gel
GF254 plates. The results of
research were tested with MANOVA (Multivariate Analysis of
Variance). From the results of present study
are known that the effect of sucrose concentration on callus
biomass and ginseng
saponin content of
Java (Talinum paniculatum Gaertn.) at
various times of
culture. The concentration
of sucrose is
right for the getting
the best biomass
is the giving
sucrose 60 g/L
at the culture
time of 6
weeks for fresh weight and 60 g/L at 8 weeks for dry weight. While the
best saponin levels produced at sucrose concentration of 90 g/L at 8 weeks of
culture.
