EKSPLORASI BAKTERI SELULOLITIK PADA TINGKAT KEMATANGAN GAMBUT YANG BERBEDA DI KAWASAN HUTAN LINDUNG GUNUNG AMBAWANG KABUPATEN KUBU RAYA
ABSTRACT: Generally, peat soil
has a low fertility rate. Based on the level of ripening, peat soil is composed
of three categories, i.e. fibric, hemic and sapric. Biologically ripening peat
soil is facilitated by cellulose degrading bacteria or cellulolytic bacteria.
This research is aimed at determining the density and obtaining the cellulolytic
bacteria genera on three levels of peat soil ripening. This research was
carried out for three months, from April to June 2014. Soil samples were taken
according to the level of ripening in the area of the Gunung Ambawang protected
forest in the District of Kubu Raya. The isolation was carried out by using the
Pour Plate method using the CMC (Carboxymethylcellulose) medium and bacteria
density calculating using the TPC (Total Plate Count) method. Observation and
characterization were made macroscopically, microscopically and through a
series of biochemical tests. The result of research was obtained five bacteria
genera, i.e. Neisseria, Actinobacillus,Acinetobacter,Staphylococcus and
Micrococcus. Most cellulolytic bacteria genera were found in the hemic soil,
whereas at least were on fibric soil. The highest rate of cellulolytic bacteria
density in the research locations was found on the sapric soil, i.e. 1,845 x
106 CFU/g, and the lowest on fibric soil, i.e. 7,8 x 105 CFU/g and the density
on hemic soil, i.e. 1,475 x 106 CFU/g.
