RESPONS PERTUMBUHAN KALUS CATHARANTHUS ROSEUS YANG DIBERI PERLAKUAN TRIPTOFAN
ABSTRAK: This research aims
are to find out the induction and growth respons of C. roseus callus on the
tryptophan treatments. Three steps have been done in this research, 1) callus
induction, 2) culture callus that treated tryptophan, 3) qualitative and
quantitative assay callus culture. The
concentration of tryptophan (ppm) that treated are 0 (T0), 100 (T1), 125 (T2),
150 (T3), 175 (T4), 200 (T5) and 225
(T6). The research results showed that the MS media for callus induction
was NAA 2 ppm and Kinetin 0,2 ppm that produced compact callus with hairy
roots. Fresh and dry weight of C. roseus callus that treated with tryptophan
decreased at 100, 125, 150 ppm and 225
ppm. However, callus that treated 175 and 200 ppm tryptophan inccreased. The
highest growth of callus occurred at callus 200 ppm (T4) with the dry weight
0,251 g. The tryptophan affected of enzymes induction and other activities for
sinthethized of alkaloid. These enzymes
correlated with decreased of the callus growth. The callus growth that treated
tryptophanm better than callus control
after 40th day subculture. The callus control growth showed necrotic and
brownees.
